Sé que desde luego significa certainly, pero no lo oigo mucho.
¿Se puede usar desde luego en lugar de vale, o de acuerdo, o sí claro, OK, bueno, como no, etc?

You can use desde luego meaning all the things you said, but it's probably more formal.
¿Puedo usar tu boli?
Desde luego.
¿ Vas a ir a ese restaurante a comer pimientos con Poli?
Desde luego.
Anyway as you said, the meaning is closer to por supuesto (certainly)
