Tu mayor fallo es que te crees que siempre llevas la razón.
Your biggest shortcoming is you always thing you're right.

shortcoming could also be replaced with defect, mistake, error, etc.

I don't want to be picky, but shortcoming isn't quite the same as error, at least not for me. Fallo = fault or error, but in the context of Tu mayor fallo es que te crees que siempre llevas la razón, fallo = fault or shortcoming,

Fault / shortcoming / weakness / defect - all suggest a continuous state of being, whereas mistake is an instantaneous thing, thus incorrect above.

In a different context, Tu mayor fallo could be your biggest mistake

Your biggest mistake was marrying that woman.

Perhaps I am overreading the difference.
