I have a bit of doubt about that Tengo un poco de duda sobre eso ¿Es necesitado "de" ahí?

Using 'poco de duda' is correct.

Could you say tengo poca duda meaning I have little doubt, i.e. the opposite?

Both are correct, but I think there is a slight nuance in meaning:
Tengo poca duda = I'm almost certain. Tengo un poco de duda = I'm not that sure.
