Fault = falta (sustantivo) error

Faltar puede ser, según el contexto:
  • To lack X (bastante formal). Falto recursos para conquistar la ciudad. I lack the resources to conquer the city.
  • To be X short to be short X. Falto un euro, ¿te pago mañana? I'm a euro short; can I pay you tomorrow?
  • To be missing X. Perdona, camarero, faltamos un tenedor. Excuse me, waiter, we're missing a fork.
  • En expresiones de tiempo, There is/are X to go. Faltan diez minutos para que el tren salga. There are ten minutes to go until the train leaves.
  • Tal vez otras frases también, pero esas son las que se me ocurren sin buscarlo en un diccionario.

Can we use lack with the preposition of after it?

Yes, but not with time. E.g. there is a distinct lack of good ideas
