¿Puedo tener una foto de tu y yo?

Is that the right way to say it in Spanish?
Are there other ways to say it?

After a preposition (like 'de') you must use prepositional pronouns. Neither tú nor yo are prepositional pronouns. Those are subject pronouns.

Your question in English seems strange to my ears.
"Me and you" isn't proper English, and I can't tell whether you're asking, "May I get someone to take a picture of you and me together?", or whether you'd like to have an extant photo given to you.

"May" is not expressed with "puedo".

What is the question you're asking?

¿Le importaría fotografiarnos juntos?
¿Le importaría tomarnos una foto a los dos?

Why are you using 'tener'. That means possessing an existing photo. You said you meant "have someone take a photo."

Julvenzor gave you perfect sentences.
You can use 'sacar' or 'tomar'.

No problem. But first I have to ask my friend if she wants a photograph taken before I ask someone to take the photograph.

Would this be ok?
