No - he referred to her as Usted because he was a humble faun in the other world, and he knew she was a princess.

But in the given example, Sancho Panther is right.
It's not a "voseo" form, is a form of address ( similar to a high-level "Usted", it's used with "usted" verbs declination).
Vos is the form fo persons with greatness.
In this example, the humble faun must speak to vos to the princess.
It's like the japanese "-san" (Usted) or "-sama" (Vos)
Very usual in medieval times. Examples
"Vos, Don Quijote, sois la persona que estaba esperando"
"Vos, gran rey, espero que me concedais este favor"

1.- form of address.
2.- "Tu" in someplaces of the world (Argentina, Bolivia, Costa Rica,.....)
