If you're trying to say: "My wife and I do the chores"

Can you say,

Mi esposa y yo hacemos quehaceres.

Yes. You can use tareas instead of quehaceres too.

It would be better if you added an article before the direct object.

Welcome to the forums, adventure. Don't know if you'd like another word for chores but I've heard them said as orden y aseo

Mi esposa y yo

...nos ocupamos de la casa.
...nos encargamos de los quehaceres domésticos.
