I know we don’t use ‘de’ with ‘thousands’:

• I think there are 2,000 people living Olivares.

• Over 500,000 people live in the city centre.

We do, however, use ‘de’ with ‘millón’:

• There are 1,000,000 people living Jerez.

• There are 8,000,000 people living London.

But if we have 'mil’ built into a number with 'millón', should we still use ‘de’?

• I think there are 1,200,000 people living Jaen.

Many thanks in advance.

No, there is no need for "de" in your last sentence. Only when the amount is expressed in terms of "millones" and not of "miles".

- Un millón seiscientos mil euros. (1,600,000)
- Tres mil millones de células. (3,000,000,000)
- Dos millones y medio de dólares. (2,500,000)

"De" is also needed for "billón" and "millardo".
