If you say "quién", you can't use "tú" for person agreement, but "él/ella".
On the other hand, "¿quién habla?", may sound a bit rude, unless there is an informal tone in the conversation. And yes, it's common to ask who wants to talk to that person, because the one answering the phone needs to pass the message. You can also ask "¿quién lo/la busca?" Informal tone: --¿Bueno? --Hola, Juan. ¿Qué estás haciendo? --¿Quién habla? --Soy Pedro. --Ah, Pedro, ¿cómo estás? ... Formal tone: --¿Bueno? --Buenos días, ¿podría hablar con la señora Marta Garrido?" --Un momento, ¿quién la busca? --La Licenciada Margarita Martínez.
