¿Hay alguna diferencia entre estas dos frases?

Sólo debería tardarme diez minutos en llegar allí a pie.
Solamente debo tardar diez minutos en caminar allí.

(It should only take me ten minutes to walk there.)

¿Se puede quitar la palabra ‘en’?

No hay ninguna diferencia de significado.
"Debería", sin embargo, expresa una ligera duda al respecto de cuánto tiempo tardaré en llegar.
"Tardarme" es un uso enfático del pronominal.

Y no, no puedes quitar "en", aunque puedes sustituirlo con "para".
