Sometimes "pues" and "entonces" are used indiscriminately.
Con este brindis, pues, termino mi discurso.
Con este brindis, entonces, termino mi discurso.
Other times, we use "pues" in an answer:
¿Fuiste allí? Pues no fui
In this case, "entonces" is not possible.
We use "entonces" when we are relating some events that have happened one after another:
El herido se levantó del suelo, entonces se puso a chillar como un loco.
"Pues" in sentences about "time" is not used, we use "entonces".
"Pues" is also used as a "pet word" (I mean a "muletilla"):
Anda, vamos, pues.
